dc.description.abstractEl objetivo de este estudio fue expresar y purificar las proteínas recombinantes (pR) Cry1Aa, Cry1Ab, Cry1Ba y Cry1Ca a partir de cepas recombinantes de Escherichia coli y evaluar su efecto sobre larvas de primer instar de Copitarsia decolora. Las pRs se evaluaron en un bioensayo preliminar sobre larvas de C. decolora a una concentración de 4,8 µg de pR/ml. Se utilizó el método de contaminación superficial sobre hojas de astromelia (Alstromelia spp). En cada tratamiento se evaluó la mortalidad y la inhibición de crecimiento basado en la longitud y peso promedio larval. La electroforesis SDS-PAGE reveló bandas de proteínas con pesos moleculares esperados para las proteínas Cry1 de aproximadamente 135 kDa. La mortalidad larval no fue considerada como variable afectada por los tratamientos debido a que fue baja (entre 0 y 10%). La pRCry1Ab mostró el mayor efecto tóxico sobre la reducción en longitud (27%) y peso (40%) promedio larval (8,90 ± 1,88 mm y 10,23 mg) comparado con los controles negativos (agua: 12,27 ± 2,68 mm y 17,90 mg; y buffer: 12,10 ± 2,66 mm y 16,40 mg). Las pRCry1Aa y pRCry1Ca fueron moderadamente tóxicas y produjeron significativa inhibición del crecimiento en larvas de C. decolora (P<0,05). El menor efecto tóxico se observó con la pRCry1Ba (12,04 ± 2,25 mm y 15,96 mg) con un 1,19 y 7% de inhibición en longitud y peso, respectivamente. Un segundo bioensayo se realizó utilizando la pRCry1Ab y evaluando cinco concentraciones. Se observó inhibición de crecimiento en la longitud (de 18,6 a 6,70%) y en el peso larval (48 a 19,8%) a medida que se aumentó la concentración de cada tratamiento (P<0,05). El porcentaje de inhibición larval en las diferentes concentraciones empleadas con Bt. HD1 y pRCry1Ab fueron estadísticamente similares (P>0,05) ya que los valores de longitud y peso se traslaparon entre los tratamientos. Es importante evaluar el efecto de las proteínas Cry recombinantes empleadas en este estudio en concentraciones superiores a 307,2 µg/ml, y otras toxinas clivadas y activadas contra larvas de C. decolora.spa

The pRs were evaluated in a preliminary bioassay against C. decolora larvae at a concentration of 4.8 μg/ml. Surface contamination method on leaves of astromelia (Alstromelia spp) was used for the bioassay. In each treatment, mortality and growth inhibition were evaluated based on the length and weight of the larva. The SDS-PAGE electrophoresis revealed protein bands with approximate molecular weights of 135 kDa. Larval mortality was not considered as an affected variable by the treatments because it was low (between 0 and 10%). The pRCry1Ab showed the greatest toxic effect on the reduction in length (27%) and weight (40%) of the larva (8,90 ± 1,88 mm and 10,23 mg) compared to the negative controls (water: 12,27 ± 2,68 mm and 17,90 mg; and buffer: 12,10 ± 2,66 mm and 16,40 mg). The pRCry1Aa and pRCry1Ca were moderately toxic and produced significant growth inhibition in C. decolora larvae (P<0,05). The lowest toxic effect on growth it was observed with the crude extract of pRCry1Ba (12,04 ± 2,25 mm and 15,96 mg) with an inhibition in length and weight of 1,19 and 7%, respectively. A second bioassay was performed evaluating five concentrations and using the crude extract of pRCry1Ab. Inhibition of larval growth in length (from 18,6 to 6,70%) and weight (from 48 to 19,8%) was observed as the concentration of each treatment was increased (P <0.05). The larval growth in different concentrations used for both Bt. HD1 and the crude extract of pRCry1Ab were statistically similar (P> 0.05) since the values of length and weight overlapped between treatments. The crude extracts of pRCry1Aa, pRCry1Ab and pRCry1Ca of Bt produced a significant inhibition of growth in larvae of C. decolora. It is important to evaluate the effect of the extracts of Cry proteins used in this study in concentrations higher than 307,2 μg/ml, also, it is recommended to determine the effect of these and other toxins cleaved and activated against C. decolora larvae.spa
